Abstract:
There are currently no drugs available that block neuronal cell death, for use in any of the wide variety of neurodegenerative diseases. Through an in vivo screening approach, we have discovered the ‘P7C3-class’ of neuroprotective aminopropyl carbazoles. We have shown that these molecules stabilize mitochondria and block neuronal cell death in multiple models of neurodegeneration, including Parkinson's disease, amyotrophic lateral sclerosis, traumatic brain injury and peripheral nerve injury. Thus far, our agents appear devoid of any signs of toxicity, despite prolonged administration and diligent pre-clinical pharmacology. We are optimizing the P7C3-class of molecules for potency and efficacy, while addressing known or perceived chemical liabilities. Our hope is that these efforts will lead to development of a drug to treat neurodegenerative disease.
