Abstract:
Telomere dysfunction has been implicated in both aging and cancer. In mammals, telomere is tightly regulated by the telomerase that mediates telomere elongation and telomere-binding proteins that protect telomere ends via complex networks of protein-protein and protein-nucleic acid interactions. Often the complexes are assembled in a transient and dynamic way to regulate cellular signaling processes. Various methods have been developed in the past to identify protein complexes, however, technologies that detect transient interactions are lacking. Here we report our recent progress to develop methods to study telomere regulatory networks, with a focus on transient and dynamic interactions. The comprehensive identification of telomere regulatory factors constitutes a key step towards therapeutic targeting of telomere dysfunction induced diseases.
