Systematic Genetic Approaches to Understand TB Pathogenesis

Biography: Dr. Sassetti received his PhD in Immunology from the University of California San Francisco and performed his postdoctoral training in mycobacterial genetics at the Harvard School of Public Health. He is currently a Professor of Microbiology and Physiological Systems at the University of Massachusetts Medical School and the Howard Hughes Medical Institute.
Abstract:The outcome of tuberculosis is influenced by genetic polymorphisms in both the host and pathogen. To investigate this complex interplay, we have systematically varied the host and bacterial genomes, and quantified the effects of these paired perturbations on disease in the mouse model of TB. The resulting "host-pathogen genetic interactions” can be used to simultaneously define protective immune pathways and the bacterial functions to resist them. Parallel genetic studies in mice and humans indicate a primary role for granulocytic inflammation in TB disease progression and suggest a variety of new interventions.
