研究方向:

我们研究1)高分辨冷冻电镜断层成像(cryo-Electron Tomography, cryo-ET)方法开发;2)新发致病型囊膜病毒的组装及通过膜融合去组装的结构与功能。

高分辨cryo-ET是通过收集维断层图像,再使用子断层图像平均法(sub-tomogram averaging, STA)进行结构解析的跨尺度结构生物学方法。由于cryo-ET数据包含精确的Z轴信息,它对样品纯度及构象均一性要求较低,且在制备样品过程中无需使其脱离原生环境,获得的结构更具生理意义。

包括新冠病毒在内的囊膜病毒是对现代人类健康最具威胁的病原体。由于形态不均一,囊膜病毒的原位结构一直是个挑战。我们使用cryo-ET解析全病毒结构,及在细胞中直接观测病毒生命活动的原位结构,希望回答以下重要问题:囊膜病毒如何组装,又如何通过膜融合去组装?可否找到病毒在这个过程中的弱点?
