研究兴趣:
生殖系细胞(包括全能性的受精卵和卵裂期胚胎细胞、多能性的内细胞团细胞和着床前后的上胚层细胞、单能性的原始生殖细胞、胚胎期生殖细胞、卵泡发生过程中的雌性生殖细胞、精子发生过程中的雄性生殖细胞、成熟卵细胞、成熟精子)是多细胞生物实现遗传信息代际传递以维持物种延续的关键类型细胞。在生殖系细胞中发生的所有表观遗传学变化都要在下一代个体的生殖系细胞中发生逆转(重编程)以恢复到跟上一代一样的表观遗传状态,因而生殖系细胞拥有最复杂也最精密的表观遗传编程和重编程机制。对生殖系发育的深入研究对于理解人类物种命运以及不孕不育相关疾病的临床诊断和治疗具有重大意义。本实验室主要围绕人类生殖系发育研究多种相关干细胞的自我更新和分化发育调控的表观遗传调控机理,以及生殖系发育的表观遗传编程和重编程机理。利用我们发展的单细胞功能基因组学高通量测序技术体系(单细胞转录组、基因组、DNA甲基化组、染色质可及性组、组蛋白修饰组、转录因子结合组、基因组三维结构组、以及多组学测序等技术),基因编辑技术、哺乳动物胚胎显微操作技术、类器官培养技术、以及人类胚胎干细胞体外定向分化等技术在单细胞和单碱基的极限分辨率下深入研究人类生殖系细胞发育以及肿瘤发生过程中基因表达网络的表观遗传学调控机理,并在此基础上深入探索生殖系细胞发育异常相关的不孕不育、以及癌症等疾病的诊断和治疗策略。
