细胞的“身份”长期被记录在染色质修饰之中,但这些化学标记是可被反复抹除-重写的,那么在不断翻新的标记体系里,稳态如何被维系?又是什么作用让系统跨过阈值、滑向疾病?为回答这一核心问题,实验室同时发展创新实验技术与计算模型,并聚焦两条互补生物体系:(1)神经细胞的转录-表观遗传稳态:神经元如何在数年乃至数十年内保持活动依赖的基因表达程序?稳态的变化如何导致阿尔茨海默病(AD)和成瘾?(2)T 细胞的命运选择:基因调控稳态的变化如何决定增殖、杀伤与耗竭之间的平衡?能否重新设定 “稳态阈值” ,以增强肿瘤免疫疗效或缓解自身免疫?
我们发展基因组分析、编辑与调控技术,多组学、多模态数据的分析框架,力求描绘出稳态-失稳的能量景观,从数据和实验中解析关键因素,寻找可逆干预窗口,为神经退行性疾病与免疫相关疾病提供新的治疗策略。
