Topic：Identification of Cross-linked Peptides from Complex Samples for Structural Analysis

Abstract

Chemical cross-linking combined with mass spectrometry (CXMS) is a powerful tool for analyses of protein structures and protein-protein interactions. We have developed a CXMS workflow, which combines a variety of readily available cross-linkers, high-resolution mass spectrometry, and a data analysis software program called pLink. This method was validated with proteins of known structures and further tested on protein complexes, crude immunoprecipitates, and whole-cell lysates. We have also extended the workflow to map protein disulfide bonds.
