许瑞明博士1964年出生于浙江天台,1984年毕业于浙江大学物理系,1989年获美国布兰迪斯大学物理学博士。现为中国科学院生物物理研究所研究员、副所长,生物大分子国家重点实验室主任。自1996年在美国冷泉港实验室独立开展研究工作以来,许瑞明博士一直致力于真核生物基因表达与调控的结构生物学研究,主要研究方向包括表观遗传调控的分子机理研究。表观遗传的核心科学问题是特定染色质结构的建立和动态调控的分子机制。许瑞明博士在染色质高级结构建立和维护的结构基础、组蛋白甲基转移酶的催化机理和活性调节以及表观遗传信息解读机制等研究方向作出了重要贡献。许瑞明2009年获“杰出青年基金”支持,2012年入选美国科学促进会Fellow。

Abstract:

Lysine acetylation is among the first post-translational modifications of histones discovered, and it plays prominent roles in chromatin-based regulation of gene expression. Enzymatic acetylation and deacetylation of histones provide major means of modulation of degrees of histone acetylation. Apart from spatiotemporal changes of enzyme abundances, conformational dynamics and protein-protein interaction can significantly influence enzymatic activities of certain histone acetylases and deacetylases. We have studied the structural basis for regulation of deacetylase activities of Sirtuins, as well as the molecular mechanism by which histone chaperone Asf1 stimulates the acetylation of lysine-56 of histone H3 – a modification critical for DNA damage response. Our studies provide mechanistic insights into allosteric regulation of histone acetylation and deacetylation, and this knowledge should benefit the design of small molecule modulators of histone/protein acetylation.
