吴虹博士1983年毕业于北京医学院,1991年获美国哈佛医学院生物化学博士。现为北京大学生命科学学院讲席,院长。
吴虹博士20余年来致力于肿瘤分子遗传学的研究,重点探讨PTEN抑癌基因在肿瘤发生中的分子机制。吴虹实验室在PTEN抑癌基因信号传递、肿瘤干细胞及基因工程动物模型等研究方面做了大量的工作,对于肿瘤发生的分子机制以及肿瘤治疗的新途径有理论和实际意义。 吴虹博士获得多项国际生物研究奖,其中包括2010年Fellow of American Association for Advancement of Science, 2016年EMBO Associate Member。
Abstract:
PTEN is the first tumor suppressor identified to have phosphatase activity and its gene is the 2nd most frequently deleted/mutated tumor suppressor genes associated with human cancers. Germline PTEN mutations are also the cause of three inherited autosomal dominant disorders. Our work demonstrated genetically that one of the major intracellular targets of PTEN is the phospholipid PIP3, the product of PI3 kinase. Additional studies from my lab further demonstrated that PTEN negatively regulates stem cell G0-G1 cell cycle entry, self-renewal, proliferation, and survival and its roles in the formation of “cancer stem cells”. We investigated the in vivo function of the PTEN and its controlled PI3K pathway in human cancer initiation and progression by generating number of genetically engineered animal models as well as PDX models. These cancer models offer unique tools for exploring both the molecular mechanisms underlying human cancers and the development of new therapies. In this seminar, I plan to discuss our recent progress in identifying the master regulator of T-ALL (acute T lymphoblastic leukemia) and how its expression determines the fate of leukemia-initiating cells.
