Pathogenesis and molecular targets of nasopharyngeal carcinoma

Biography:1998年毕业于中山医科大学,获博士学位,曾在美国Tufts-New England Medical Center做博士后。主要从事鼻咽癌发病机制及靶标研究。主持国家重点研发计划,国自然重点项目等国家级项目或课题多项,以通讯作者在《J Clin Invest》、《PNAS》、《JNCI》、《Nat Commun》、《PLoS Pathog》、《Cancer Res》等发表SCI收录论文50多篇。
Abstract:Nasopharyngeal carcinoma (NPC) is the most common head and neck cancer in South China and Southeast Asia. Epstein-Barr virus (EBV) is implicated as an etiological factor in nasopharyngeal carcinoma. However, the mechanisms of cell-free EBV infection and transformation of nasopharyngeal epithelial cells (NEPCs) remain elusive. We found Bmi-1 is major oncogene in NPC. Bmi-1 alone could induce immortalization and EMT in NEPCs. The Bmi-1 induced premalignant NPECs-Bmi1 cells grown as sphere-like cells (SLCs) presented significantly enhanced EBV infection. By using this cellular infection model, we identified non-muscle myosin heavy chain IIA (NMHCIIA) and neuropilin 1 (NRP1) as potential cellular factors for EBV infection of NPECs. Furthermore, by RNA-seq we identified several potential molecular targets such as FGFR3-TACC3 and RARS-MAD1L1 in NPC, which might enable the practice of novel targeting therapy in NPC.
